How to use sybr safe dna dye

The following is taken from the SOLABOR technical article.

GelRed is a highly sensitive, low-toxic and super-safe fluorescent nucleic acid gel staining reagent. It can replace ethidium bromide (EB), and its sensitivity is much higher than EB, and it does not need decoloration. Because GelRed and EB have the same spectral characteristics, it is not necessary to change the imaging system when replacing EB with Gelred. GelRed can be used for preformed gel and dyeing after electrophoresis. Generally, staining after electrophoresis is more sensitive and can eliminate the interference of dyes on DNA migration. After electrophoresis, it is dyed with GelRed, which is simple to operate and does not need decoloration and special solution. As long as the dye is diluted in 0. 1M NaCl and the gel is put into the dyeing solution, it can be observed after 30 minutes. The dye solution is very stable at room temperature and can be reused. Comparatively speaking, the preformed gel does not need additional dyeing process, so the dosage of dye is less, which is simpler and more economical.

Characteristics of gelled nucleic acid dyes;

Gelred's water-soluble dye has passed the safety certification of the US Environmental Protection Agency, and the waste can be directly poured into the sewer without causing any environmental pollution.

Important note:

1, non-toxic: GelRed's unique oiliness and high molecular weight prevent it from penetrating cell membrane and entering cells. Ames test results also show that the mutagenicity of this dye is much less than EB. Its water-soluble dye has passed the safety certification of the US Environmental Protection Agency, and the waste can be directly poured into the sewer without causing any environmental pollution.

2. high sensitivity: it is suitable for electrophoresis staining of fragments of various sizes, and has less influence on nucleic acid migration than sybr green I. ..

3. High stability: it can be heated by microwave oven and stored at room temperature.

4. High signal-to-noise ratio: the sample has strong fluorescence signal and low background signal.

5. simple operation: like EB, there is no need to worry about dye degradation during pre-forming and electrophoresis; After electrophoresis, the dyeing process only takes 30 minutes, without decoloration or washing, and can be directly observed by ultraviolet gel transmission instrument.

6. Wide application range: suitable for pre-gel (gel staining method) and post-gel electrophoresis staining (bubble staining method).

7. Perfect compatibility with standard gel imaging system: When using 3 12nm excited UV gel imaging system, GelRed can perfectly replace EB.
