1. Purpose
To standardize the use and maintenance of Shimadzu LC-10AT high-performance liquid chromatography system.
2. Scope
This procedure applies to the use and maintenance of Shimadzu LC-10AT high-performance liquid chromatography system.
3. Responsibilities
The instrument analyst in the quality inspection room is responsible for the daily use and maintenance of the Shimadzu LC-10AT high-performance liquid chromatography system.
4. Procedure
4.1 System composition: This system consists of 2 LC-10ATvp solvent delivery pumps (main/A pump and deputy/B pump), Rheodyne 7725i manual injection valve, and SPD-10Avp UV-visible detection It is composed of processor, SCL-10AVP system controller, CLASS-VP (Ver. 6.1) workstation and IBM desktop computer.
4.2 Preparation
4.2.1 Before use, the required mobile phase should be prepared according to the test method of the sample to be tested, filtered with a suitable 0.45μm filter membrane, and degassed ultrasonically for 20 minutes.
4.2.2 Replace the appropriate chromatographic column (the position of the column inlet and outlet should be consistent with the flow direction of the mobile phase) and quantitative loop according to the needs of the sample to be tested.
4.2.3 Prepare sample and standard solutions (can also be prepared when balancing the system), and filter them with a suitable 0.45μm filter membrane.
4.2.4 Check whether the power cord, data cable and infusion pipeline of each component of the instrument are properly connected.
4.3 Start up
4.3.1 Turn on the power supply, turn on the uninterruptible power supply, pump B, pump A, detector, and system controller in sequence, and wait until the self-test of the pump and detector is completed. Then, turn on the computer monitor and host.
4.3.2 Start the CLASS-VP workstation
4.3.2.1 After confirming that the instrument and system controller are started, turn on the computer power and select Start > Programs > from the Windows taskbar Chromatography > CLASS-VP to open the CLASS-VP menu window. (You can quickly open the CLASS-VP main menu through the shortcut on the desktop)
4.3.2.2 Double-click the Instrument icon in the Main Menu window.
4.3.2.3 Enter the username and password to log in to CLASS-VP. By default, the user name and password are "system" and "2001" respectively.
4.3.2.4 When SCL-10Avp beeps, it means that the connection with the computer has been established, and the Instrument window opens.
4.3.2.5 Starting the Chromatogram Monitor
4.3.2.5.1 Select Program > Chromatography > Chromatogram Monitor from the Windows taskbar to open the Chromatogram Monitor window.
4.3.2.5.2 Set the type and format of monitored items as needed.
4.3.2.6 Starting Offline Processing
4.3.2.6.1 Double-click the Offline Processing icon in the Main Menu window.
4.3.2.6.2 Double-click the instrument name set up for data collection to open the instrument for offline processing.
4.4 CLASS-VP initial system settings (setting method default parameters)
4.4.1 Click the "New" button, and then click New Method to initialize the method parameters.
4.4.2 Select commands from the Method menu of the Option tab, and change the parameters of each command in the dialog box to select commonly used options.
4.4.3 Select File > Method > Save as Default to save the options set here as default parameters.
4.5 Set analysis parameters
4.5.1 Set LC parameters
4.5.1.1 When setting new analysis parameters, please click the "New" button Select New Method in the open list, the default method parameters will be loaded, and the method file name "Untitled.met" will be displayed in the LC Setup Assistant window. The existing method file can be opened by clicking the Open button.
