Project introduction:
This project focuses on the key technical problems of bioreactor, and adopts vegetable oil system to realize the efficient expression of foreign protein and solve the problem of protein separation and purification. Based on the theory of molecular chaperone and recombinant protein expression, the soluble expression system of foreign protein was constructed, and the soluble and efficient expression of foreign protein was realized. The problems of easy degradation, instability and short half-life of exogenous drug proteins were solved by site-specific chemical modification technology.
Taking advantage of the advantages of strong ability to express foreign protein, short growth cycle and easy large-scale production, a technical platform for expressing foreign protein by edible fungi was established. Using the secretion and expression system of Escherichia coli, the target protein is secreted into the periplasm of cells, which reduces the purification steps, reduces the production cost and makes the product quality more controllable.
Fibroblast growth factor (FGFs) and growth hormone expressed by the key technology of bioreactor were used to develop freeze-dried powder, eye drops, injections, gels and body intervention preparations for national second-class medical devices. At present, the above drugs and machinery have entered clinical trials to design large-area burns, chronic refractory wounds, chronic granulation wounds, wound healing after plastic surgery, growth hormone deficiency and so on.
The project applied for national invention patent 1 1, and granted 8 items. In recent three years, the cumulative output value of patented technology products has reached 1 1. 5.8 billion yuan, with remarkable economic and social benefits.
