After virus samples are collected, PCR testing is usually not possible at the sample collection site in a timely manner, so the collected virus swab samples need to be transported, and the virus itself will soon be cleaved outside the body, affecting subsequent testing. For detection, it is necessary to add virus preservation solution during storage and transportation. Currently, it is mainly divided into two types: inactivated type and non-inactivated type.
For different detection purposes, different virus preservation solutions need to be used. The two currently widely used preservation solutions have their own characteristics. In order to meet different testing requirements and different virus detection laboratory conditions, it is very necessary to sample different preservation solutions.
Inactivation preservation solution: It is mainly a virus lysis preservation solution modified from nucleic acid extraction lysis solution. It contains a high concentration of guanidinium salt during sampling, which can effectively inactivate the virus and effectively prevent the operator from being infected again. However, it also contains RNase inhibitors, which can protect the viral nucleic acid from being degraded, so that it can be subsequently detected by NT-PCR. . And it can be stored at room temperature for a relatively long time, saving the cost of virus sample storage and transportation.
Non-inactivated storage solution: It is mainly a virus maintenance solution based on the transport medium. It can maintain the activity of the virus in vitro and the integrity of the antigen and nucleic acid. It contains Hanks solution base, gentamicin, fungal antibiotics, BSA (V), cryoprotectants, biological buffers and amino acids, etc., to protect the viral protein shell. Decompose and maintain the originality of virus samples to a large extent. In addition to being used for nucleic acid extraction and detection, this preservation solution can also be used for virus culture, isolation, and antigen detection. However, long-term storage after sampling requires strict low temperature.
